2226. ANA-profile (BlotAnalisis)

The ANA Profile (immunoblot analysis) is an extended test for antinuclear antibodies (ANA) used in the diagnosis of autoimmune diseases, including systemic lupus erythematosus (SLE), scleroderma, and Sjögren’s and Raynaud’s syndromes.

ANA immunoblot analysis is recommended in cases of a positive ANA IFT result (antinuclear factor detected by indirect immunofluorescence testing), which indicates the presence of antinuclear antibodies in the serum. The detection of ANA is associated with autoimmune processes; however, a negative result does not exclude the presence of disease.

The ANA Profile is an extended laboratory test that detects IgG autoantibodies to nuclear and cytoplasmic antigens associated with systemic autoimmune diseases.

The comprehensive panel includes testing for IgG antibodies to 16 of the most clinically significant antigens involved in the development of systemic connective tissue diseases:

• nRNP+Sm, Sm, RNP (70, A, C), SS-A, SS-B, Ro-52, Scl-70, PM-Scl, Jo-1, CENP B, PCNA, dsDNA, nucleosomes, histones, ribosomal P proteins, AMA-M2.

Immunoblot analysis allows the detection of specific ANA in blood serum, which may occur either in combination or individually. Some ANA subtypes are associated with a particular disease or with overlap syndromes.

To distinguish transient induction of autoantibodies from their persistent presence in an autoimmune disease, repeat testing may be necessary.

It should be taken into account that antinuclear antibody results obtained at different laboratories may vary due to differences in the methods and antigens used.

• For screening for autoimmune diseases in patients with fever of unknown origin, arthritis, or skin rashes.
